Il naît d’une famille aisée à Chinon. En 1532, il commence à rédiger son célèbre livre « Pantagruel » sous le pseudonyme Alcofribas Nasier. Il cherche à déjouer la censure, à cause de son personnage et d’une satire trop ouverte. Quelques années après, il suit Joachim du Bellay en Italie en tant que médecin. Dès son retour en France, il entame la suite de « Pantagruel », avec « Gargantua », puis un troisième ouvrage en lien à ces deux personnages : « Tiers Livre ». Enfin, entrecoupé de voyages à Rome, il rédigera « Quart Livre » qui lui vaudra une sévère censure.
